In observing the recent use of ulex europaeus agglutinin-I (UEA-I) as a marker of human vascular endothelium, we investigated the existence of lectins that can be a marker of cerebrovascular endothelium in several common laboratory experimental animals, using both light and electron microscopy. As lectins, concanavalin A (Con A), UEA-I-HRP, and peroxidase conjugate of wheat germ agglutinin (WGA-HRP) were used. The brain tissues of man, monkey, cat, dog, rabbit, Wistar rat, and Mongolian gerbil were collected. The rabbit, rat and gerbil were observed to positively react with Con A in almost all the intracerebral blood vessels. On the other hand, WGA-HRP was definitely positive on the vascular endothelium of all the animals. Almost all the blood vessels in human brain tissues were clearly stained with UEA-I-HRP. On electron microscopic examination, WGA binding sites were limited to the luminal plasma membrane of vascular endothelium. As a conclusion appropriately selected types of lectins, i. e. WGA for animals and UEA-I for the human brain according to the present experiment, are apparently useful as a very fine marker of vascular endothelial cell membrane in many animal species.
