An experiment was carried out to clarify details in changes related to angiospasm of Renin granules contained in the juxtaglomerular apparatus (JGA), the intrarenal blood distribution, and the vasoconstriction rate (VCR) of perinuclear vacuoles in intrarenal media cells. In it 15 Wistar rats were used. In each rat 5 ml of blood was removed, Noradrenaline injected, and the renal arterial or renal arteriovenous clamping performed. Observation was made on changes in Renin granules of the JGA, the VCR of the intrarenal arteries, and the intrarenal blood distribution. It was done in rats 10 minutes after the appearance of hypovolemic shock and Noradrenaline injection, as well as in rats 30 minutes after the renal arterial or renal arteriovenous clamping. It was possible to draw the following conclusions from the experiment. (1) When the intrarenal blood distribution was observed, cortical ischemia and medullary congestion were present in the group of rats showing hypovolemic shock. Cortical congestion and medullary ischemia were found in the group of rats injected with Noradrenaline. Both cortex and medulla were ischemic in the group of renal arterial clamping, and congestion in the group of renal arteriovenous clamping. (2) The VCR was high in both lobar and interlobular arteries in the groups of hypovolemic shock and renal arterial and renal arteriovenous clamping. It was remarkable high in the lobar artery and low in the interlobular artery in the group of Noradrenaline injection. (3) There was a clear relationship between hypovolemic shock and intrarenal angiospasm. The pathological changes of the nephrons under both conditions were characterized by those of Renin granules in the JGA of the kidney. (4) As compared with Renin granules in the physiological condition, these granules decreased in the group of hypovolemic shock and increased in the group of Noradrenaline injection. This is because Noradrenaline injection caused the condition of the lobar artery which led to an increase in VCR. Renin granules decreased in the group of renal arterial clamping, probably because there was a decrease in the amount of circulating blood in the kidney and severe angiospasm occurred to the lobar and interlobular arteries. Renin granules also decreased in the group of renal arteriovenous clamping, probably because there was no blood circulation in the kidney, in spite of an increase in the amount of intrarenal blood and because severe angiospasm occurred to the lobar and interlobular arteries. (5) These results indicate that the appearance of perinuclear vacuoles in the media of intrarenal blood vessels was a morphological evidence for the occurrence of angiospasm. Changes in Renin granules in the JGA depended upon the degree of VCR, the site of vasoconstriction, and the amount of circulating blood in the kidney.
