This study was performed in order to clarify morphometrically the reinnervation process of the muscular branch nerve (SOL nerve) and intramuscular nerves (IM nerve) of the soleus, which are the most peripheral portions of the severed sciatic nerve, for up to 60 weeks, compared with a control group of 24 week-old rats. A comparison of the regenerated fibers of the SOL nerve with that of the IM nerve elucidated the effects of two different manipulations of the severed sciatic nerve : (1) entubulation of the stumps utilizing a silicone tube, and (2) wrapping the stumps with collagen film. Both sides of the SOL nerve and IM nerve of 52 rats were examined by electron microscopy at 1,2,3,4,6,8,12,16,24,36,and 60 weeks after the operation. The findings of reinnervation of these nerves were observed. Morphometric analysis in the diameter, the thickness of the myelin sheath and the density of regenerated myelinated nerve fibers were measured on electron microscope photographs. The major findings are summarized as follows ; Reinnervating nerve fibers were reduced in thickness of fibers and myelin sheath, and significantly increased in number of the fibers ; however the increased numbers were limited to the proximal portion, up to the fascicles of classes 3 and 4,and significantly reduced in number to almost the same value as the controls in the fascicles of classes 1 and 2 of the IM nerve, the most distal portion of the peripheral nerve in both the tube and film groups. These findings of the classes 1 and 2 appeared from about 12 weeks to 60 weeks after the operation. The results suggest that in order to improve recovery from the damages, the manipulations of the severed stumps are important, but clarifying the efficiency and utility of the factors affecting nerve growth in the periphery in the muscle tissue might be much more important.
