A further analysis of the immune mechanisms responsible for the induction of experimental autoimmune hepatitis in C57BL/6 mice was performed by a syngeneic assay system using sensitized spleen cells and isolated hepatocytes. The sensitized nylon wool adherent fraction showed a highest cytotoxic activity compared to other fractions, and this activity was markedly decreased by the treatment with anti-Thy 1, 2 antibody and complement. The cytotoxic activity of this fraction was increased after depletion of B-cells and Fc receptor bearing T-cells, although it was slightly decreased after further depletion of macrophages. No cytotoxic activity was found in this fraction against syngeneic glomerular epithelial cells and EL-4 cells, and anti-liver cell membrane antibodies could not be detected in the culture media nor on the surface of recovered target hepatocytes obtained after incubation of target and effector cells. These results suggest that cytotoxic T-cells are mainly responsible for the induction of experimental hepatitis in C57BL/6 mice.
