Cloning of Glucose Phosphorylating Genes in S. cerevisiae by the KUMethod and Application to ATP Production

Genes of hexokinase (HK) and glucokinase (GLK) of Saccharomyces cerevisiae were cloned onto vector plasmid YEp13 and the hybrid plasmids obtained were called pYHl and PYGI. Selection was done after transformation by the KU-method using intact yeast cells treated with alkali cations. Dried yeast cells prepared from the strain DKD-5D-Hharboring the hybrid plasmid pYHl or PYGI phosphorylated glucose about 3 times as effectively as dried cells of the strain without plasmid. These cells with pYHl or PYGI completely converted adenosine (130 him) into adenosine 5-triphosphate (ATP) (130 him), consuming glucose rapidly by the enhanced glycolytic pathway, while the strain without plasmids could not.
