Metabolic Transit of Lysinoalanine in the Rat Body: Observation of Feeding Tests with Alkali-treated Casein and Gluten

Gluten and casein were dissolved in 0.2n NaOHat 40°C for 2 hr. The alkali-treated proteins were partially hydrolyzed with Pronase E, prior to preparing diets for feeding tests on rats. The diet based on the alkali-treated gluten hydrolysate and those based on the alkali-treated casein hydrolysate contained lysonoalanine (LAL) at levels of 1 7.5 μmol/g and 30.8 μmol/g, respectively. After the administration of the diets, the rats were sacrificed and investigated for the transport of LAL. In both cases, a major part of LALentered the portal vein without being hydrolyzed to free LALand eventually reached the kidney. The bound LALin the kidney was chromatographically diverse, lying in a molecular weight range of 300-500 daltons. A significant amount of free LAL was produced from the bound LALby treatment with Pronase E. This indicates that, when rats were given a hydrolysate of gluten or casein treated under the present conditions, low-molecular peptide-bound LALoccurs to be transported to the kidney.
