In our continuing study on intracellular signal transduction mechanisms using pharmacological probes of natural origin, we have been searching for the lipophilic low-molecular-weight compounds that can easily pass through the cell membrane and have some effects on signal transduction steps. The purpose of the present study is to isolate the active compounds from South American medicinal plants that have an influence on the expression of genes for interleukin-2 (IL-2), PEBP2 α A, a transcription factor playing a critical role in osteoblastic differentiation, and osteoclastogenesis-inhibitory factor (OCIF). The dichloromethane extract of Brazilian medicinal plant, Canela de cheiro, Octea opifera (Lauraceae) showed a potent enhancement activity of IL-2 gene expression and two naphthoquinones (1, 2) were isolated from the active fraction. Also three novel sesterterpenoids (3-5) and four new xanthones (6-9) were isolated from the whole plant of Peruvian folk medicine, Hercampuri, Gentianella alborosea and G. nitida (Gentianaceae). Furthermore, ten cucurbitacin derivatives including four new compounds (10-19) were isolated from the fruit of Brazilian medicinal plant, Buchinha, Luffa operculata (Cucurbitaceae). Their structures were elucidated by means of spectral methods, especially using two dimentional NMR experiments. A novel sesterterpenoid (3) is the frist isolation of a seco-type sesterterpenoid with the new ring skeleton. Two new cucurbitacin derivatives (10, 11) are the first example of cucurbitacins having the six-membered lactone on A ring. Seven compounds (1-5, 7, 8) were examined the enhancement activity of IL-2 gene expression and ten cucurbitacin derivatives (10-19) were tested inhibition activity of PEBP2 α A and OCIF gene expression together with morphological change activity. As a result, three compounds (2, 5, 7) showed enhancement activity of IL-2 gene expression. Five cucurbitacins (10, 12-15) showed inhibition activity of PEBP2 α A and OCIF gene expression and almost all cell shape were change to round.
