Significance of LDL binding Lymphocytes in Patients with Hyperlipidemia

Low density lipoprotein (LDL) affects some immunological lymphocyte functions and also controls cholesterol metabolism through binding with LDL receptors. A decrease in LDL receptor activity has been reported in familial hypercholesterolemia. However, it is not clear whether B or T lymphocytes possess LDL receptors. The previous method using isotope labelled LDL did not indicate which lymphocytes are responsible for the decrease in the LDL receptor in familial hypercholesterolemia. In this study, we employed FITC-conjugated LDL for the receptor assay. This approach could differentiate LDL-binding lymphocytes clearly from lymphocytes not bearing LDL. These LDL-binding lymphocytes showed the characteristics of B cells. A B cell rich population carried LDL, while a T cell rich population carried little LDL. The majority of LDL-binding cells collected by a fluorescence activated cell sorter (FACS II) bore immunoglobulins on their cell surface. In clinical application, the percentage of LDL-binding lymphocytes in hyperhpidemic patients was significantly lower than that in healthy adults (P<0.01). These results indicate that the decrease in LDL receptor activity in hyperlipidemic patients is not due to a decrease in the number and/or the affinity of receptors but to a decrease in the number of LDL-binding lymphocytes. This method using FITC-labelled LDL may be applicable in the purification of LDL-binding cells, and may be useful for studies on the effects of LDL on immunological functions and lipid metabolism in lymphocytes.
