Dissociation of Digoxin-Antibody Complex after Addition of Dextran-Coated Charcoal in the Radioimmunoassay of Digoxin

In radioimmunoassay of digoxin using dextran-coated charcoal, longer charcoal adsorption time resulted in less radioactivity in supernate which contains antibody-bound digoxin. Since the quantitation of digoxin by radioimmunoassay is dependent on the degree of radioactivity in the supernate, the above-mentioned effect is not desirable for obtaining reliable results. The results obtained from this study indicated that the decrease of radioactivity in supernate may be due to the cumulative effects of a continuous dissociation of digoxin-antibody complex after the addition of coated charcoal and of adsorption of anti-digoxin antibody on the charcoal. These effects were intensified at higher temperatures. Cooling of the reagents is recommended to minimize the error caused by such effects.
