Studies on Optimal Conditions for the Determination of Human Immunoglobulin-Secreting Cells by a Protein A-Reverse Haemolytic Plaque Assay

Pokeweed mitogen (PWM)-stimulated immunoglobulin-secreting cells (ISC) were examined in a reverse haemolytic plaque assay (RHPA) using protein A-coated sheep red blood cells (SRBC) as indicator cells. The range of concentretions of 5-40μl/ml PWM was optimal in generating ISC. Peak response in the kinetics of PWM-stimulated immunoglobulin (Ig)-secreting cells was consistent after 6-8 days under the optimal culture conditions. Using this method, culture of PWM-stimulated peripheral blood lymphocytes (PBL) from normal donors produced a mean of 16, 280 (±537) IgG plaque forming cells (PFC) per 10^5 lymphocytes, 4,910 (±267) IgM PFC/10^5 and 2,910 (±223) IgA PFC/10^5 (±standard deviation). This method was more sensitive than the conventional method and showed relatively favorable reproducibility. Accordingly, this plaque assay was a useful and sensitive in vitro test for the evaluation of polyclonal B-cell activation in humans, and might provide additional informations with reference to study on disturbance in the Ig synthesis by lymphocytes from patients with various disorders.
