Anti-cow's Milk IgE Antibodies Recognize Fewer Molecules than IgG Antibodies

A variety of allergic disorders following ingestion of cow's milk have been reported. In some cases, IgE mechanisms have been established, but when IgG antibodies to cow's milk antigens are present in sufficient quantities the immunologic mechanisms leading to allergies remain unclear. In order to clarify the relationship between IgG and IgE antibodies against cow's milk antigens, the molecules these antibodies recognize were analyzed by radioimmunoassay and Western blotting using native and digested cow's milk antigens. In radioimmunoassay, IgG antibodies reacted with digested cow's milk antigens, but no IgE antibodies to digested antigens were detected. In Western blotting, IgG antibodies reacted with bovine serum proteins, casein, β-lactoglobulin and α-lactalbumin. IgE antibodies reacted with bovine serum proteins and casein but not with α-lactalbumin or β-lactoglobulin. Neither IgG nor IgE antibodies bound to the low molecular weight digested cow's milk antigens (less than 14,000). These results show that anti-cow's milk IgE antibodies recognize fewer molecules than IgG antibodies.
