Sandwich Enzyme Immunoassay for Serum Amyloid A Protein in Cerebrospinal Fluid

Serum amyloid A (SAA), a putative precursor of the AA protein which constitutes amyloid fibrils in secondary amyloidosis, has been clinically evaluated as a sensitive acute-phase reactant in serum. At present, the SAA concentration in serum is analyzed automatically by the latex nephelometry, but this assay does not detect low concentrations of SAA because of low sensitivity. An enzyme immunoassay (EIA) for determining low concentrations of SAA is described. The method is a sandwich immunoassay using purified rat anti-AA monoclonal antibody (coating antibody), rabbit polyclonal antibody to human SAA and peroxidase conjugated anti-rabbit γ globulin. The assay is sensitive, reproducible and reliable and does not require pretreatment of the specimen prior to assay. In this study, we attempted to apply our EIA method for the measurement of SAA in cerebrospinal fluid (CSF) of patients with various types of meningitis.
