Pharmacological properties of traditional medicine (XXXI) : Anxiolytic effect of Hangeshashinto and the combinations of its major constituents on rat experimental colitis

半夏瀉心湯(HST)は日本の伝統的漢方医学において精神神経症で悪化する炎症性潰瘍性消化管疾患を治療するための漢方処方として使用される。前報でHSTが2, 4, 6-trinitrobenzene sulfonic acid (TNBS)の直腸注入で誘導される大腸炎を改善したことを報告した。本研究では, 同様のラット大腸炎モデルにおけるHSTの抗不安効果を明らかにした。ラットの不安行動は高架式十字迷路(EPM)で測定した。TNBS注腸によりラットは不安様行動を示した。HSTを経口投与したラットのEPMにおける行動観察では, 不安様行動の改善効果を示した。活性成分を同定するためにberberine (BE), baicalin (BA), glycyrrhizin (GL), 人参サポニン分画(GS)について検討した。それら4成分を同時に経口投与することにより, HSTの抗不安効果の約半分の効果が見られた。また, 前報で大腸炎を改善したGLとGSを同時投与したときには効果は見られなかったが, BAとBEの併用では効果が見られた。以上の結果から, TNBS大腸炎ラットにおいてHSTには抗不安効果と大腸炎改善効果があり, その効果は部分的にそれぞれBAとBE, GLとGSの複合効果によると考えられた。
