<Short Communications>Effect of PMS+VC on the Activities of Na^+, K^+-ATPase, K^+-Dependent Phosphatase and Phosphorylation of Intermediate

Na^+, K^+-ATPase preparation was prepared by the 2 M Nal treatment from guinea pig kidney microsomes. The effects of PMS and vitamine C on the Na^+, K^+-ATPase activity and K^+-dependent phosphatase activity in the Na^+, K^+-ATPase membrane preparation were investigated. PMS alone (10^<-8>-10^<-4>M) had no effect on both enzyme activities but inhibited both enzyme activities when 5 mM vitamine C was added. Vitamine C showed biphasic effects on the enzyme activities. Vitamine C alone inhibited Na^+, K^+-ATPase at low concentration (10^<-5>-10^<-4>M) but did not show inhibition at lower and at higher concentrations. When added with 10^-5 M PMS, vitamine C showed inhibitions on the enzyme activity at high concentration (10^<-4>-10^<-2>M). The addition of PMS and vitamine C also inhibited the formation of phosphorylated intermediate. When the enzyme preparation was treated with PMS and VC and assayed for both enzyme activities and phosphorylation simultaneously, the order of inhibition became like below : Na^+, K^+-ATPase>K^+-dependent phosphatase>>phosphorylation of the enzyme. The presence of K^+ is favorable for the inhibition of PMS+VC on Na^+, K^+-ATPase activity and K^+-dependent phosphatase activity. Mg^<2+> and Na^+ antagonized the action of K^+. Chelating agent, EDTA destroyed the inhibition of PMS+VC even in the presence of K^+. Then it is suggested that the inhibition of PMS+VC occurred on E_2 state of the enzyme and, heavy metals are necessary for the inhibiting process of PMS+VC. These inhibitions of PMS+VC on the enzyme activities were found in the membrane preparations from kidney, brain and heart of guinea pigs.
