Effects of Rb^+ and Choline^+ Substitution for K^+ on the Growth of HeLa Cells

The cell proliferation, cellular protein content and intracellular contents of K^+, Rb^+ and Na^+ were investigated in HeLa cells cultivated in the different culture media supplemented with calf serum. When a half of K^+ in the medium was replaced by Rb^+, the cell growth was not significantly inhibited, but protein content slightly decreased. Nearly half K^+ in the cells was exchanged by Rb^+. Protein synthesis as observed from the protein content and the cell growth were repressed in the medium, in which 90% of K^+ was replaced by Rb^+. Intracellular K^+ content decreased to less than 15%, but the Rb^+ content became the same with the normal K^+ content. On the other hand, a replacement of 90% of K^+ by choline^+ did not significantly affect intracellular K^+. The cell growth and protein synthesis were maintained normally. Intracellular Na^+ content was not influenced by the medium exchange. Thus, intracellular K^+ seemed to be an essential requirement for the cell growth and protein synthesis, but extracellular K^+ did not affect directly. Rb^+ was not as effective as K^+ to support the cell growth.
