The Na^+, K^+-ATPase activities in the brain microsomal membrane were studied in the stroke-prone spontaneously hypertensive rats (SHRSP) and control normotensive Kyoto-Wistar rats (KW). SHRSP showed significantly higher level of the Na^+, K^+-ATPase activity than KW at 5-7 months old. But the characteristics of Na^+, K^+-ATPase activities were demonstrated to be very similar between SHRSP and KW. The dose response curves of Na^+ , K^+-ATPase activities on various concentrations of Mg^<2+>, Na^+ and K^+ were studied. SHRSP showed increased V_<max>, but very similar K_m for these cations as compared with KW. Both SHRSP and KW showed almost the same profiles when dose response curves were expressed by the percentages of maximal levels. The ouabain inhibition of Na^+, K^+-ATPase activities also gave the same profiles between SHRSP and KW. However, the ouabain binding to Na^+, K^+-ATPase increased in SHRSP as the increased activities of the enzyme in SHRSP. Then it is suggested from these results that increased numbers of the enzymes and sodium pumps are involved in the membrane of the brain cell of SHRSP, though there is no difference in qualitative characteristics of the enzyme between SHRSP and KW.
