<Short Communications>Effects of Chelaters and Metal Ions on the Inhibition of K^+-Dependent Phosphatase by PMS+VC

Effects of chelating agent and some metal ions were studied on the inhibition of PMS (Phenazine methosulfate)+VC (Vitamine C) on K^+-dependent phosphatase activity in 2 M Nal treated microsome preparation from kidney of guinea pig. The inhibition of PMS+VC on K^+-dependent phosphatase was almost completely antagonized by EGTA and o-phenanthroline as well as EDTA. Then some metal ions were assumed to be necessary for the inhibition. Mg^<2+> did not increase the inhibition of PMS+VC but decrease it. CaCl_2,MnCl_2,ZnCl_2 and CuCl_2 inhibited K^+-dependent phosphatase activities in the presence of PMS+VC, but caused stronger inhibitions on the activities in the absence of PMS+VC. FeCl_2 also showed similar effects but FeCl_3 increased the inhibition of PMS+VC without inhibition of control activity. The ferric ion increased maximum inhibition of PMS+VC. It is suggested that iron ions may be important role on the inhibition of PMS+VC on K^+-dependent phosphatase activity and Na^+, K^+-ATPase activity.
