Effects on Cultured Cells of Increased Concentration of Potassium in Combination with Ouabain : Contents of Na and K, ATP Level and Cell Volume in Mouse L Cells

The present studies demonstrated the effects of higher concentration of extracellular K ([K_o]) on L 929 cells in combination with ouabain. Intracellular Na (Na_i) increased and extracellular K (K_i) decreased after 3 h treatment by 0.05-0.8 mM ouabain, but the value of Na_i+K_i was almost constant. Similarly, K_i rose up and Na_i fell, as [K_o] increased from 5.5 to 85 mM. When some combinations of [K_o] and ouabain, e.g. 10.5 mM and 0.05 mM, respectively, were used, K_i was similar to that of control. After 24 h treatment with high [K_o], K_i increased and the cell growth was suppressed, whether ouabain was added or not. Na_i rose up only when ouabain was applied. This suggests that K_i seems to have a closer relation to the cell proliferation than Na_i, and that there is a difference in sensitivity to ouabain between K and Na pump in the cell membrane. Ouabain, or high [K_o] except 85 mM, scarcely affects the cellular ATP level. The dicline of the cell volume by increasing [K_o] became less significant as ouabain concentration increased (0.5-0.8 mM).
