Effects of Ouabain on the Growth, Internal Concentrations of Na^+ and K^+, and Volume of Mouse L Cells

Studies were made on the effects of ouabain on L 929 cells. The cell growth was not influenced by 0.1-0.2 mM ouabain, whereas it was suppressed by 0.4-1.6 mM ouabain. The cells could not proliferate after 48 h in the presence of 0.8-1.6 mM ouabain. Protein synthesis was also inhibited by 0.3 mM ouabain. Intracellular concentration of Na^+ was increased but that of K^+ was decreased in 1.5 h by 0.3 mM ouabain, and the concentrations were kept unchanged after that time. The sum of those concentrations did not alter during the treatment with ouabain. Similar results were obtained when varied concentrations of the inhibitor were used for 3 h. Intracellular Na^+ increased and K^+ decreased in the concentration range of ouabain less than 0.2 mM, and they approached to certain levels when ouabain concentration exceeded the range. A decrease in cell volume, which was dependent upon ouabain concentration, occurred in parallel with the decrease in intracellular K^+. ATP-level, respiration and production of lactic acid were not influenced by 0.3 mM ouabain.
