Studies on Simplified Procedures for Extraction and Sensitive Assay of Adenine Nucleotides in Cultured Mammalian Cells

Extraction of adenine nucleotides was completed within 2 minutes, when water suspensions of L929 cells were incubated in hot water. Concentrations of nucleotides did not alter during a 20 minutes incubation. Hot water treatment for more than 1 minutes was sufficient to inactivate ATP hydrolyzing enzymes in the cells, but that for less than 30 seconds was not. Also, enzymes used for the conversion of ADP and AMP into ATP were inactivated by 5 minutes incubation in hot water, hence 7 to 10 minutes was the minimum and sufficient time to extract the nucleotides and to stop the enzymatic conversions. Hot water was a more reliable extractant of adenine nucleotides than hot isotonic NaCl and Na_2SO_4,and hot PBS, or cold PCA. A glass mixing apparatus was designed, by which the assay of 10^<-4> to 10^<-8> M ATP can be made within 10 seconds by using luciferin-luciferase solutions. Thus, ATP assays of more than 30 samples become capable within 1 hour.
