Effects of PMS (Phenazine methosulfate)+NADH on Na^+, K^+-ATPase activity and K^+-dependent p-nitrophenylphosphatase (K^+-PNPase) activity were studied on the NaI treated microsomes prepared from guinea pig kidney. 10^<-5> M PMS+10^<-3> M NADH destroyed both Na^+, K^+-ATPase and K^+-dependent phosphatase activities. NADH alone had no effect on both enzyme activities. The inhibitory potency by PMS+NADH is due to reduction, because it was demonstrated that PMS+NAD had no effect on Na^+, K^+-ATPase and K^+-dependent phosphatase activities, and the inhibiting ability of PMS+NADH remained on an anoxic condition. The inhibition process of PMS+NADH may contains heavy metals because the inhibition by PMS+NADH was shown to be antagonized by a chelating agent, EDTA.
