Effects of vanadate on Na^+, K^+-ATPase and K^+-dependent p-nitrophenylphosphatase activities were compared by using 2 M NaI treated microsomes from kidneys of guinea pigs and the effects of ligands on these inhibitions were investigated. The both enzyme activities were inhibited by very low concentrations of vanadate but the activities of Na^+, K^+-ATPase were one order less sensitive to vanadate than the K^+-dependent phosphatase activities. The presence of potassium ions and magnesium ions was necessary for the inhibition by vanadate of Na^+-, K^+-ATPase activity but was not so much necessary for the inhibition of K^+-dependent phosphatase activity. Sodium ions antagonized the inhibition of K^+-dependent phosphatase by vanadate. ATP reduced the inhibitory action of vanadate on Na^+, K^+-ATPase activity but p-nitrophenylphosphate did not reverse the inhibitory action of vanadate on K^+-dependent p-nitrophenylphosphatase activity. When optimal concentrations of MgCl_2 (25 mM) and KCl (25 mM) for the vanadate inhibition were used, the vanadate sensitivity of Na^+, K^+-ATPase increased and came near the sensitivity of K^+-dependent phosphatase.
