[^3H] ouabain binding to Na^+, K^+-ATPase in guinea pig kidney membrane preparations was investigated. The ouabain binding did not occur significantly on the conditions of Mg^<2+>, Mg^<2+> and Na^+, and Mg^<2+>, Na^+ and K^+. The ouabain binding occurred in the presence of ATP, Mg^<2+> and Na^+, and ATP, Mg^<2+>, Na^+ and K^+. The treatment of PMS (Phenazine methosulfate)+VC (vitamine C) decreased the association rate of the ouabain bindings to Na^+, K^+-ATPase, but did not affect optimal binding levels. The inhibition by PMS+VC on K^+-dependent phosphatase was examined in the presence of various concentrations of ouabain. The apparent Ki of ouabain was not affected significantly by PMS+VC and the inhibition by PMS+VC was not affected significantly by the concentrations of ouabain.
