Na^+, K^+-ATPase and K^+-dependent-phosphatase activities in Na^+, K^+-ATPase preparation from the microsome of guinea pig kidney were inhibited potently by quercetin. The 50% inhibition of both enzyme activities occurred at about 50 μM quercetin. Mg^<2+>-ATPase activity was not inhibited so much as Na^+, K^+-ATPase activity. The enzyme activities in crude microsomes and Nal treated microsemes were similarly inhibited. Quercetin inhibited K^+-dependent phosphatase activity by decreasing maximal activity and K^+ affinity to the enzyme. Mg^<2+> decreased quercetin affinity to the enzyme. Quercetin inhibition of Na^+, K^+-ATPase was suggested to occur, by the same mechanism, on K^+-dependent dephosphorylation step of the ATPase reaction.
