Na^+, K^+-ATPase activity in kidney microsomal membrane was studied in the stroke-prone spontaneously hypertensive rats (SHRSP) and control normotensive Kyoto-Wistar rats (KW). SHRSP showed lower level of the Na^+, K^+-ATPase activity than KW at 3-7 months old though the difference was insignificant. The effects of various concentrations of sodium and potassium ions were also investigated. SHRSP showed decreased V_<max>, but same K_m with KW. Both SHRSP and KW showed the same profile when dose response curves were expressed by the percentages of maximal level. It is suggested from these results that decreased activity of kidney membrane Na^+, K^+-ATPase might be related to Na^+ retention and the pathogenesis of hypertension of SHRSP.
