A new apparatus was developed, which was available for assaying the respiration of cultured cells either attaching to or suspended in a commercial plastic culture dish. The apparatus was equipped with an oxygen electrode and a magnetic mixing rotor. The rate of cell respiration was calculated from the slope at the linear part of time course curves of the polarographic current. The calculation was made on a basis that one per cent reduction in the current corresponded to 2.47μM oxygen consumption according to the calibration curve of this oxygen electrode. The respiratory rate seemed to be higher in suspended cells than in attaching cells obtained from replicate cultures, but the difference between the two rates was not statistically significant. The coefficient of variation in the rate was higher in attaching cells than in suspended cells. As far as the mixing rotor was operated at 60 rpm, the medium mixing neither detached adhering cells nor produced injury effect on cell morphology, as shown by protein assay and microscopic observation, respectively.
