Agonistic or antagonistic action of Kampo medicines used for menopausal symptoms on estrogen receptor subtypes, ERα and ERβ

これまでに我々は婦人科系疾患に用いられる漢方薬がエストロゲン様活性を有することを明らかにした(和漢医薬学雑誌22巻4号228-236(2005))。本研究では更年期関連疾患に頻用される8処方と既報告で最も高いエストロゲン様活性を示した葛根湯の計9処方について,エストロゲン受容体(ER)α及びβに対するアゴニストあるいはアンタゴニスト作用を解析した。ERへの結合解析には受容体/コファククー・スクリーニングシステムを用いた。その結果ERα及びERβに対してアゴニスト作用を有する処方:TJ-1。ERβに対してアゴニスト作用を有する処方:TJ-7,TJ-24,TJ-25,TJ-61。ERα及びERβに対してアンタゴニスト作用を有する処方:TJ-15,TJ-67。ERβに対してアンタゴニスト作用を有する処方:TJ-23,TJ-106であった。本研究においては,われわれは漢方薬がERに直接結合することを初めて見出した。また9処方のいずれもERβへの作用が認められたことから,更年期関連疾患に頻用される漢方薬は,主にERβを介して薬理作用を示す可能性があることが示唆された。
