Maoto, a Kampo medicine, suppresses human serum-induced motility of human breast cancer cells

• われわれは,麻黄湯を経口投与したマウスの血清が高転移性マウス骨肉腫細胞の運動能を抑制すること,また,がん転移モデルマウスを用いた解析で,麻黄湯の経口投与により肝転移が抑制されることを初めて見出した。麻黄湯を臨床応用するためには,麻黄湯がヒトのがん細胞に対して作用するのかどうかを明らかにする必要がある。そこで本研究では,ヒト血清を用いたヒトがん細胞の運動能測定法を新たに確立し,ヒト血清により誘導されるヒトがん細胞の運動能に対する麻黄湯の効果について解析した。ヒト乳がん細胞株MDA-MB-231細胞の運動能は,ヒト血清濃度依存的に誘導され,1%血清で最も強く誘導された。この血清濃度では,細胞障害作用を示さなかったことから,運動能測定法には1%ヒト血清を用いることにした。この測定法を用いて,ヒトがん細胞の運動能に対する漢方薬の効果を解析した。麻黄湯は添加濃度依存的にMDA-MB-231細胞の運動能を抑制し,100μg/mlの濃度で83%抑制した。しかし,比較方剤として用いた十全大補湯と四君子湯は,がん細胞の運動能に影響を与えなかった。また,6.25μg/mlから100μg/mlの濃度において,麻黄湯はがん細胞の増殖に影響を与えなかったことから,麻黄湯による運動能の抑制は,細胞障害によるものではないことが明らかになった。これらの結果から,麻黄湯がマウスのがん細胞だけでなく,ヒトのがん細胞の運動能も抑制することが明らかになり,麻黄湯は,ヒトのがん転移を抑制する可能性があることが示唆された。
