The antitussive activity and site of antitussive action of 1-[2-{α-(p-chlorophenyl)benzyloxy}ethyl]piperidine hydrochloride(HT-11) were investigated. The antitussive activity of HT-11 was 0.78 times and 1.36 times that of codeine in dogs and cats, respectively. The site of action seemed to be the cough center per se. This conclusion was derived from the analyses of the actions as follows : (1) Comparison of equi-active dosages in four different routes of administration. (2) Effects on the sensory receptors in the tracheal mucosa and on the pulmonary stretch receptor. (3) The influence of decerebration. (4) Effects on the centrally-induced cough response and on the centrally-induced sustained inspiratory response. (5) Effect on the efferent respiratory pathways. In addition to the above results, the drug showed strong antihistamine and papaverine-like actions on the tracheal muscles.
