Rabbits were intravenously administered 10mg/kg salicylic acid with or without 20 mg/kg phenylbutazone. After the drug administration, plasma protein binding of salicylic acid was examined using a semi-microultrafiltration method, and concentrations of salicylic acid and its metabolites in the plasma and urine were determined using the GLC method. The binding of salicylic acid to plasma protein was significantly decreased after the administration of phenylbutazone. It was assumed that salicylic acid competes with phenylbutazone for the same binding sites. However, the reduction of plasma concentration of salicylic acid due to the competitive displacement was shown at the initial stage after the simultaneous administration. Furthermore, remarkable depression of salicylate elimination from the rabbit plasma was observed and this may be attributable to the inhibition of renal excretion of salicylic acid by phenylbutazone. From the finding that simultaneously administered phenylbutazone increases the unbound ratio of salicylic acid and yet keeps the plasma concentration of salicylic acid significantly high, it can be speculated that the salicylate distribution into the tissues might be further enhanced.
