Studies on the Formation of Vitamins and Their Functions in Hydrocarbon Fermentation : (II) The Forms of Vitamin B_<12> Produced by Several Kinds of Bacteria Grown in Hydrocarbon Media

This paper deals with the form of vitamin B_<12> produced in the cells of bacteria grown in a medium containing hydrocarbon as a sole carbon source. The results obtained are follows : 1. The amount of the vitamin produced in the hydrocarbon medium was considerably more than in the carbohydrate media under the experimental conditions. 2. The greater part of the vitamin produced in the hydrocarbon medium was retained in the bacterial cells, while most of the vitamin was extruded into the cultural filtrate in the case of the carbohydrate media. 3. The B_<12>-vitamers formed in the cells of Corynebacterium simplex were extracted with hot alcohol and purified by ion-exchange cellulose column chromatography. The identity of the main component with 5,6-dimethylbenzimidazolylcobamide coenzyme (DBCC) was demonstrated by paper chromatography, paper ionophoresis, alsorption spectrophotometry, assay of its coenzyme activity in diol dehydratase system and other tests. 4. The vitamin B_<12>-active substance formed in the cells of other hydrocarbon-utilizing bacteria, such as Nocardia gardneri, Mycobacterium smegmatis also showed a similar paper chromatographic behavior with DBCC. 5. On the basis of these results it can be concluded that vitamin B_<12> produced in the hydrocarbon medium exisist mainly as its coenzyme form in the bacterial cells. The possible role of the coenzyme in hydrocarbon-metabolism was also discussed.
