Fractionation of Tryptophan from Polypepton Effective for Bacterial β-Amylase Production : Studies on Microbial β-Amylases (X)

Among four fractions of Polypepton (fractions A, B, C and D) obtained by gelfiltration of Sephadex G-25,fraction D was found to enhance β-amylase production when added to the basal medium containing fraction B. By paper chromatography, amino acid analysis, and UV and IR spectral analysis, fraction D was identified as tryptophan. The addition of L-tryptophan in place of fraction D showed almost the same effect on the enzyme production. There was little difference in effects on the enzyme production between the L- and DL-forms of tryptophan. However, it should be noted that only tryptophan among twenty amino acids tested could stimulate the enzyme production in the presence of fraction B.
