In order to construct a tetracycline (Tc) inducible expression-secretion vector system, the α-amylase structural gene of Bacillus subtilis strain 1A412 was isolated, and inserted between the promoter-operator and the terminator region of a Tc-resistant (Tc^r) gene. When the α-amylase gene was expressed in B. subtilis on a plasmid, the expression of the α-amylase gene could be modulated by the Tc concentration. α-Amylase activity was increased approximately 15-fold by adding 500 ng/ml Tc.
