A mathematical technique is proposed to optimize the feed rate of the growth-limiting substrate for metabolite production by fed-batch culture. The technique aims to maximize one of the objective functions, i.e. the yield or productivity, under various constraints. The unique structure of the mathematical model that describes the fed-batch culture system allows the use of a simple analytical procedure based on Pontryagin's maximum principle in solving the optimal changes in the specific growth rate and in the total amount of biomass. In this technique, when the feed rate of the substrate solution calculated along the optimal change in the specific growth rate is negative, then the optimal policy must be replaced by a suboptimal one. A simple "suboptimal" policy is proposed, i.e. "first a batch operation then the operation along the originally optimal change in the specific growth rate". A numerical example is presented to demonstrate how the optimization technique is applied.
