Repeated Use of Lipase Immobilized on Amphiphilic Gel for Hydrolysis of a Small Amount of Glycerides Included in Liquid Crude Fatty Acid

The water content of wet amphiphilic gels such as Octyl-Sepharose CL-4B and Phenyl-Sepoharose CL-4B was about 94%. The buffered (0.1M phosphate buffer, pH 7.0) gels adsorbed above 97% of lipase dissolved. They were easily dispersed in liquid crude fatty acid. The immobilized lipase was used to hydrolyze glycerides five times repeatedly in a reaction system of the disperseed gel in the liquid crude fatty acid (glyceride content, ca. 8%). The immobilized lipqse lost its hydrolysis activity gradually. That is to say, achievement of chemical equilibrium took a longer time and the degree of hydrolysis decreased in the successive trials. The inactivated gels were regenerated in a packed column first by eluting out both residual fatty acid around the gel particles and the accumulated glycerol with ethanol and then with water, and finally by being rebuffered with 0.1 M phosphate buffer, pH 7.0. The immobilized lipase on the regenerated gel showed the same hydrolysis activity as the original one.
