Fed-batch cultures of Bacillus magaterium AJ 3355 which produces β-amylase extracellularly were tried with several feeding modes of soluble starch into peptone-meneral salts medium. Fed-batch cultures with two kinds of feeding modes, i.e. constant feed rates and exponentially increased feed rates, gave almost the same enzyme yield when potimally operated. A yield of the enzyme of 3.6 times that in the batch culture was obtained by an improved feeding mode which was a modification of exponential fed-butch culture. The effect of feeding of the carbon sources on the enzyme biosynthesis by the bacterium was discussed in connection with the 'catabolite repression' phenomenon.A convenient laboratory-scale equipment named an automatic curve tracer was developed with which the feed rate of a substrate solution could be regulated to a predetermined pattern.
