Active Monomer of Sweet Potato β-Amylasel : Stabilizatin and an Improvdde Preparation Method Using α-Cyclodextrin

The active monomer of sweet potato β-amylase prepared by modificatio with periodate-oxidized maltohexaose was a labile molecule, but the stability of the monomer was strongly enhenced at pHs lower than 5.0 and higher than 9.0,and at 60℃ in the presence of a-cyclodextrin. The active monomer was prepared by modification with the oxidized maltohexaose in the presence of α-cyclodextrin, the resulting yield of the monomer was about 50% by weight, and the specific activity was 2,700 units per mg protein, while that of the orignal enzyme was 3,000 units/mg. Both the yield and specific activity of the monomer were much improved compared with those of the active monomer prepared in the absence of α-cyclodextrin. The stabilization with α-cyclodextrin was also observed for the original enzyme.
