Mechanism for the Formation of 2,3-Butanediol Stereoisomers in Klebsiella pneumoniae

The mechanism of the formation of 2,3-butanediol isomers in Klebsiella pneumoniae was studied. A new model is proposed with the existence of three 2,3-butanediol dehydrogenases : meso-butanediol dehydrogenase (D-acetoin forming), meso-butanediol dehydrogenase (L-acetoin forming), and L-butanediol dehydrogenase. The butanediol dehydrogenases were separated individually and their stereospecificities were determined by the use of all the pure isomers of 2,3-butanediol (D(-), L(+), and meso), acetoin (D(-), and L(+)), and the separation and determination of these isomers. Results obtained from activity staining of their enzyme bands with disc electrophoretic procedures were consistent with their stereospecificities obtained above. The presence of acetoin or butanediol racemase was not confirmed in our experiments.
