Laboratory-Scale Production of Acetoin Isomers (D(-) and L(+)) by Bacterial Fermentation

Preparation of two isomers of acetoin (D(-) and L(+)) by bacterial fermentation were studied. The D(-)-isomer was prepared from glucose by using Pseudomonas sp.S-4,which produced acetoin but not 2,3-butanediol because of the lack of 2,3-butanediol dehydrogenase. Although attempts to find favorable bacteria to produce the L(+)-isomer failed, it could nevertheless be prepared from L(+)-2,3-butanediol by using Brevibacterium saccharolyticum C-1012,which possessed L(+)-2,3-butanediol dehydrogenase but not racemase for acetoin.The isomers obtained were almost pure as judged by optical rotation measurement, NMR with a chiral lanthanide shift reagent, and gas chromatographic analysis. The yiedld of D(-) and L(+)-isomer were about 0.79 and 1.43 g per l of broth, respectively.
