Laboratory-Scale Production of 2,3-Butanediol Isomers (D(-), L(+), and Meso) by Bacterial Fermentations

Laboratory-scale production of three isomers of 2,3-butanediol (D(-), L(+), and meso) by bacterial fermentations were studied. The organisms used were Serratia marcescens IAM 1022 for preparation of the meso-isomer and Baciluus polymyxa IAM 1189 for the D(-)-isomer. Although many attempts to find bacteria having the ability to produce the pure L(+)-isomer failed, it could be prepared from the commercially available racemic 2,3-butanediol by usng Bacillus cereus IAM 1072 which was unable to oxidize the L(+)-isomer. The organism was further shown to assimilate the D(-) and meso-isomers but not the L(+)-isomer.Individual 2,3-butanediols produced were extracted with ether from the fermentation liquids, and purified by fractional distillation. The isomers obtained (D(-) : [α]^<25>_D-12.7,L(+) : [α]^<25>_D+13.0 and meso : [α]^^25__D0.0) were about 98-100% pure compared with the data of their synthetic preparations on gas chromatography and optical rotation measurements. The yields of D(-), L(+), and meso isomers were about 6.0,1.6,and 21.2 g per litre of broth, respectively.
