Effect of Ionic Strength on Optimum pH of Immobilized Trypsin on a Copolymer of 2-Acryloylamino-2-Methylpropanesulfonic Acid and Acrylic Acid

Trysin was covalently immobilized on 2-acryloylamino-2-methylpropanesulfonic acid-acrylic acid copolymer. The immobilized enzyme showed a specific activity of 65.7% of the original activity. The pH-activity profiles of the immobilized enzyme were shifted towards the alkali side, depending on the ionic strength. At high ionic strength, the activities of soluble and immobilized trypsin decreased, however, the decrease was depressed by immobilization. The optimum pH of the immobilized enzyme was raised about 2.2 pH units at the ionic strength of 3.13×10^<-2>. The optimum temperature for the immobilized trypsin was decreased by 20℃ compared to the soluble form. The immobilized trypsin activity dropped to the extent of 23% in the initial 2 weeks, and thereafter the activity remained almost the same till the end of 6 weeks.
