The structures of eicosapentaenoic acid (EPA) derivatives formed during accelerated stability testing of EPA ethylester

エイコサペンタエン酸(EPA)の腫瘍増殖に対する報告は多数ある。EPAは非常に不安定であることはよく知られていることから, 加速試験において, EPAエチルエステル(原料物質)から生じた副産物(EPA誘導体, EPADと略)の抗腫瘍効果が元のEPAエチルエステルよりも低下することが考えられた。しかしながら, EPAエチルエステル処理したルイス肺癌(LLC)細胞でのO_2^-産生は, EPAD処理したLLC細胞と比較すると同じであった。我々は, 相対湿度60±5%, 温度37℃, 30日間保存条件下で, 未反応のEPAエチルエステル(原料物質), EPA(遊離体)以外に2種類のEPAエチルエステル副産物(EPA誘導体, E-1およびE-2)を得た。E-1およびE-2は, ^1H-, ^<13>C-NMRおよびMSスペクトル解析から, 各々EPAエチルエステルの2量体およびEPAヒドロキエチルエステルであると同定した。E-1およびE-2はLLCに対するDNA合成阻害作用は, EPAエチルエステル(原料物質)よりも強かった。EPAエチルエステル, E-1およびE-2は0.03から3.3μg/mLの濃度において, LLC細胞からのO_2^-産生を増加し, E-1のO_2^-産生効果はEPAエチルエステルよりも強かった。E-1およびE-2のマトリゲル誘導血管新生に対する阻害効果はEPAエチルエステルよりも強かった。これらの実験事実は, 加速試験によってEPAエチルエステル(原料物質)から生じたEPAエチルエステル2量体およびEPAハイドロキシエチルエステルがEPAエチルエステルと同様に抗腫瘍剤の候補化合物の一つであることを示唆している。
