The Nucleotide Sequence of the β-Glucosidase Gene from Cellvibrio gilvus

In the above report, we presented the nucleotide sequence of the β-glucosidase gene isolated from Cellvibrio gilvus and compared its deduced amino acid sequence with those of the same enzyme from other microorganisms.We reported that the N-terminal half of our enzyme had sequences that were conserved in other enzymes, whereas the C-terminal had no conserved sequence. However, Henrissat (J. Ferment.Bioeng., 76,240-242,1993) has pointed out that our C-terminal half did have a conserved region too if a frame shift could be postulated somewhere around the midpoint of our reported sequence.In the light of this, we reexamined our original data and carried out an additional sequencing experiment. The overall sequencing strategy is shown in Fig. 1. As a result, delecton of two successive bases, C and G, was found between ^<1419>C and ^<1420>T in our reported sequence. The corrected nucleotide sequence with the deduced amino acid sequence is shown in Fig. 2. The corrected open reading frame has 2259 bp, and the deduced molecular weight of the coded enzyme was caluculated to be 79,941 daltons. G+C contents of 1st, 2nd and 3rd letters of the codons used were calculated to be 68.7,49.6 and 89.6%, respectively, which is in accordance with other G+C rich genes. The content of 4 amino acids (pro, Ala, Arg, and Gly) in our enzyme was found to be 37.1%. Figure 2 also shows the alignment of the amino acid sequence of our enzyme with those of same enzyme of defferent origins. Clearly, conserved sequences are found out not only in the N-terminal but also in the C-terminal region. The β-glucosidase cloned from C. gilvus is thus a typical β-glucosidase of the BGB group according to the classificaiton of Beguin et al. (Ann. Rev. Microbiol., 44,219-248,1990).
