Clinical pharmacology of ibopamine. (2). Pharmacokinetics after single and multiple oral administration in healthy Japanese subjects.

Pharmacokinetic characteristics of ibopamine were studied in healthy Japanese adult male volunteers. They received ibopamine -25, 50, 100, and200mg- in single-dose study and 100 mg three times daily for 7 days in multiple-dose study. Pharmacokinetics of ibopamine were assessed by measuring plasma and urine concentrations of metabolites (free epinine, total epinine, DOPAC, and HVA).<BR>Ibopamine itself was not determined because it was considered to be rapidly disintegrated to epinine after absorption.<BR>The total epinine was measured after treatment with perchloric acid because most of epinine in blood exists as the conjugated form, which produces epinine by hydrolysis with perchloric acid.<BR>The plasma concentration of free epinine reached the peak level within 1hr after the oral administration of ibopamine. This fact suggests that ibopamine is rapidly absorbed after oral administration.<BR>The plasma concentrations of total epinine, DOPAC, and HVA reached peak levels within 2.6 hr after administration. Among the metabolites examined, the concentration of total epinine was highest and that of free epinine was lowest in plasma after the admin istration of ibopamine. The elimination half-life of total epinine was estimated to be about 2hr. The total epinine consisted of epinine 3-O-sulfate (74-75%) and epinine 4-O-sulfate (22-24%).<BR>The plasma concentration and the urinary excretion of total epinine increased as the dose of ibopamine was increased from 25mg to 200mg, but the increase was slight when the dose was increased from 100mg to 200mg. This result suggests that the sulfoconjugation of epinine is saturated in high-dose administration of ibopamine.<BR>Neither accumulation nor significant change of pharmacokinetics was observed in the plasma levels of metabolites even after multiple administration.<BR>It was concluded that between Japanese subjects and Caucasians, pharmacokinetic characteristics of ibopamine were not so different.
