The Changes of Intramuscular Oxygen Tension by Local Cold Application in the Rat

We investigated the changes of skeletal intramuscular partial oxygen tension (Po_2) quantitatively during cold application to hind leg in rats. Intramuscular Po_2 was measured by the sensor that was a Clark polarographic cell with thermocouple, which was inserted in the calf muscle of ten male Fisher rats, 54-58 weeks of age. During the leg was cooled for 30min by applying ice packs, intramuscular Po_2 and temperature were measured. Intramuscular temperature was maximally reduced after 30min of cooling to 25 degrees Celsius. Intramuscular Po_2 was decreased gradually during cooling. Those recoveries were started after removing the cold application. However the recovery of Po_2 was slower than that of the temperature, talking approximately 30min to recover to initial conditions. These findings suggested that the cryotherapy such as ice pack could reduce the intramuscular Po_2 and keep the condition of low levels for certain periods. So excess cooling or long period-cooling could damage the hemodynamics and subsequently the function of the normal tissues.
