Effect of Hyaluronic Acid on Proteoglycan Metabolism in Cultured Bovine Articular Chondrocytes

In order to study the mechanism of action of hyaluronic acid (HA) in the treatment of inflammatory and non-inflammatory joint diseases, we have investigated the affinity of fluorescence-labeled HA to human osteoarthritic cartilage explants in vitro, and the effects of rooster comb-and Streptococcus zooepidemicus-derived HA on the synthesis and release of proteoglycan (PG) by adult bovine articular chondrocytes in vitro. Labeled HA spread in the cultured cartilage in a time-dependent fashion, and HA was preferentially accumulated in the pericellular region, where PG was lost as examined by toluidine blue staining of the cartilage. Exogenous HA (10-1000μg/ml) did not significantly affect PG synthesis as determined by ^<35>SC_4-incorporation into PG. In contrast, HA inhibited ^<35>SO_4-prelabeled PG release from the cell layer into the culture medium in a concentration-dependent fashion. A pronounced inhibition was obtained at 1000 μg/ml. These observations suggest that HA has chondroprotective effects through preserving PG in the articular cartilage matrix and, in this way, exerts a beneficial clinical effect in the treatment of these joint diseases.
