The level of cathepsin L in gingival crevicular fluid during orthodontic tooth movement and effect of tension-force on cathepsin L activity from periodontal ligament cells

The purposes of this study were (1) to investigate the levels of cathepsin L (CAL) in gingival crevicular fluid (GCF) after the start of orthodontic treatment, (2) to measure the activity of cathepsin L from PDL cells by mechanical stress in vitro. The study included 10 patients, each having one treatment tooth undergoing orthodontic movement and the contralateral and antagonistic control tooth. The GCF was sampled at the control and antagonistic sites and treatment (compression) sites before activation and at 1, 24, and 168h. Retention of plaque-induced inflammation allowed this study to focus on the dynamics of mechanically stimulated CAL level in GCF. Further, we examined the effect of tension-force on CAL activity from human periodontal ligament (PDL) cells in vitro. The GCF levels of CAL for the treatment teeth were significantly higher than the control teeth at 24h. At the contralateral and antagonistic control sites, CAL level did not change significantly over time. It was shown that tension-force significantly stimulated the activity of CAL in PDL cells. This stimulatory effect exhibited time-and magnitude-dependency in vitro. These results suggested that the amount of CAL in the GCF increased by orthodontic tooth movement and that the increase of CAL may be involved in extracellular matrix degradation response to mechanical stress.
