Electrophoretic Studies on Plant Protoplasts II . Relative Amounts of Various Charged Groups on the Surface of Barley Mesophyll Protoplasts

• Electrophoretic mobilities of barley mesophyll cell protoplasts modified by chemical and enzymatic treatments were measured in media at various pH values to elucidate the contribution of phosphate, carboxylate and amino groups to the surface charge density. Existence of these charged groups was confirmed by results of treatment of protoplasts with glutaraldehyde (for amino groups), acid phosphatase (for phosphate groups) and 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide together with glycine methyl ester (for carboxylate groups). The relative amounts of these groups were estimated from the curves of surface charge density (σ) vs. surface pH (pH_s) of the treated protoplasts, in terms of simplified acid-base dissociation curves. The estimated ratio of the amounts of phosphate, carboxylate and amino groups was approximate 0.5 : 0.5 : 0.7 for the native (unmodified) barley mesophyll cell protoplasts, when the total negative charge on the native protoplasts was assumed to be 1.
