Tetrazolium Salt Method Coupled with a Phenazinium Salt Gives Higher Rates for Dehydrogenase Reactions

Reactions of dehydrogenase were measured by the following two methods : the increase in absorbance at 340 nm of NADH (ε=6.22 mM^<-1> cm^<-1>) and the increase in absorbance at 570 nm of the formazan (ε=13 mM^<-1> cm^<-1>) formed by the 5-ethyl-phenazinium ethyl sulfate-coupled reduction with NADH of 3-(4', 5'-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide. The tetrazolium salt method gave higher rates and smaller K_m values for NAD in the reactions with alcohol, lactate and malate dehydrogenases.
