Production of DNA by Continous Culture Using Pseudomonas aeruginosa KYU-1

Pseudomonas aeruginosa KYU-1 is capable of producing a large amount of intracellular DNA as well as extracellular DNA. The accumulation of intracellular DNA was found to be growth associated. To produce DNA efficiently, we have investigated the culturing conditions of this microorganisms in continous culture and it was found that the partial pressure of oxygen in the culture liquid was the most important operation parameter influencing DNA accumulation. When the partial pressure of oxygen in the culture liquid was above 0.1atm, the biological oxygen transfer rate (biological γ_<ab>) and the specific growth rate (μ) decreased with increases in the partial pressure of oxygen, although the DNA yield from glucose remained constant. When the partial pressure of oxygen was approximately 0.1atm, the specific growth rate reached its maximum value, with the maximum DNA production rate being 0.14g/lh.
